connect the hot to one lug and the ground to another on the ohms setting....

No way to measure output unless you could somehow measure the minute currents generated by the strings over the pickup. With a multimeter you can measure the coil resistance - but this does not equate to output. For example if you put a weak (or no) magnet in the same pickup the multimeter reading will be the same but the voltage output less.. thus the guitar will drive the amp less and the sound softer. Thus the output is a function of the magnet strength and the DC resistance and topology of the coil and magnets.
